Overview
Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) based on a fragment of adrenocorticotropic hormone (ACTH 4-10) with a C-terminal Pro-Gly-Pro tripeptide extension that enhances stability and potency. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and has been approved as a pharmaceutical in Russia and Ukraine since the 1990s for nootropic and neuroprotective indications.
Mechanism of Action
Semax modulates multiple neurotrophic factors including brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and tropomyosin receptor kinase B (TrkB) signaling. It influences the expression of over 24 genes related to neurotrophic support, immune modulation, and vascular function. Unlike ACTH, semax has no steroidogenic activity and does not affect cortisol levels.
